The new flights to Orlando will operate every Tuesday and Sunday from January to March 2020.
Cook will offer two cabins on these flights with economy seats offering a seat pitch of 31 inches and premium seats with a 35-inch pitch.
Return flights start at £299.99 for bookings made by 21 June or from £319.99 after this date.
Prices include hand luggage and a three-course James Martin in-flight meal.
Seven-night packages start from £535 based on two adults and two children sharing, departing 7 January 2020.
